摘要
目的 :用自体的人肝癌细胞系诱导对肝癌细胞具有特异性杀伤作用的T淋巴细胞。方法 :分离肝癌患者外周血淋巴细胞 (PBL) ,在 5 0U/mlIL - 2存在下与经照射的自体肝癌细胞系共刺激 ,进行筛选性的混合淋巴细胞肿瘤细胞培养 (MLTC) ,直接免疫荧光法分析其细胞表型 ,51Cr释放试验检测效应细胞的杀伤效应。结果 :淋巴细胞明显增殖 ,细胞变大呈豆芽状。细胞毒性实验证明该细胞对HCC - 972 4有特异性杀伤作用 ,而对K5 6 2细胞没有杀伤作用。结论 :利用同体肝癌细胞系HCC - 972 4作为刺激细胞反复诱导肝癌患者能产生肝癌特异性的CTL。推测在刺激及培养条件适宜时 ,人外周血中较少的CTL前体细胞 (CTLp)
Objective:To generate tumor specific cytotoxic T lymphocytes from autologous human hepatic carcinoma cell line HCC-9724.Method:Peripheral blood lYmphocytes(PBL)were isolated from patients with hepatic carcinoma.Screening mixed lymphocyte tumor cell culture(MLTC)was performed by simultaneously stimulating PBL and irradiated autologous hepatic carcinoma cell lines in the presence of 50 u/ml IL-2.Phenotype was analyzed by direct immunofluorescence using flow cytometer. 51 Cr release assay was used to detect the cytotoxicity of lymphocytes.Result:After MLTC,proliferation of lymphocytes was remarkable and lymphocytes became larger like the bean sprouts.Cytotoxicity test demonstrated that the lymphocytes had specific cytotoxicity againhst HCC-9724 but had no cytotoxicity against K562.Conclusion:Hepatic carcinoma-specific cytotoxic T lymphocytes can be obtained using autologous human hepatic carcinoma cell line HCC-9724 as stimulating cells.
