摘要
目的 :观察脂质体转染反义 PKCα对 CNE- 2 Z细胞生长的影响。方法 :用脂质体法转染反义 PKCα,以 MTT法测细胞生长。结果 :(1)脂质体浓度为 2 .0 0 0~ 8.0 0 0 mg· L- 1时 ,随着脂质体浓度增加 ,细胞生长指数逐渐降低 (P<0 .0 5 ) ;但当浓度为 1.0 0 0 mg·L- 1时 ,其对细胞生长似无影响。 (2 )在 4.0 0 0~ 16 .0 0 0 mg· L- 1的范围内增加转染的反义PKCα的浓度 ,细胞生长指数逐渐降低 (P<0 .0 0 1)。结论 :小剂量脂质体转染反义 PKCα可抑制 CNE- 2 Z生长 ,反义PKCα的剂量与细胞生长抑制呈量效关系。
Objective:To observe the effect of protein kinase C-α(PKCα)antisense oligonucleotides transfected by cationic liposome(LP)on growth of CNE-2Z cells.Methods:PKCα antisense oligonucleotides were transfected by LP,and the cell growth was detected by MTT colorimetric assay.Results:(1)Cell growth index of CNE-2Z cells decreased as the strength of LP increased in the range of 2.000~8.000 mg·L -1 (P<0.05),but there seemed to be no effect on cell growth with 1.000 mg·L -1 of LP.(2)Cell growth index reduced with a concentration increase ranging from 4.000 mg·L -1 to 16.000 mg·L -1 in antisense PKCα transfected by LP (P<0.001).Conclusion:It is indicated that antisense PKCα transfected with lower dose of LP may suppress growth of CNE-2Z cells,and this inhibitory effect shows a dose-effect relationship.
