摘要
目的 :观察槲皮素对 Hela细胞细胞周期和 RNA聚合酶活性的影响及其相互关系。方法 :分别用 4、2 0和 10 0 μmol/ L 槲皮素处理培养的 Hela细胞 3d后 ,应用流式细胞仪进行细胞周期分析 ,并通过体外核转录实验测定总 RNA聚合酶活性和 RNA聚合酶 活性。结果 :Hela细胞经槲皮素处理后 ,细胞周期中 G1 期细胞增加并呈现量 -效关系 ,显示槲皮素有阻断 Hela细胞周期于 G1 期作用 ;总 RNA聚合酶活性和 RNA聚合酶 活性均受抑制 ,并呈现量 -效关系。结论 :槲皮素阻断 Hela细胞周期于 G1 期与抑制 RNA聚合酶活性有关。
Objective:To study the effects of quercetin on cell cycle and RNA polymerase in Hela cells.Methods:Hela cells were incubated with 4、20 and 100 μ mol/L quercetin for 3 days,respectively.Cell cycle was analyzed with flow cytometry,and the activities of total RNA polymerase and RNA polymerase Ⅱ were measured in isolated Hela cell nuclei by nuclear transcription in vitro.Results:After incubation with quercetin,the percentage of Hela cells at G 1 phase increased in a dose-dependent manner,and the activities of total RNA polymerase and RNA polymerase Ⅱ were inhibited in the same way.Conclusion:Quercetin is able to arrest cell cycle progression of Hela cells at G 1 phase,which may be related to the inhibition of the activities of total RNA polymerase and RNA polymerase Ⅱ.
