摘要
目的 :探讨白细胞介素 - 1α(IL - 1α)、低密度脂蛋白 (LDL)、高密度脂蛋白 (HDL)对血管细胞粘附分子 - 1(VCAM -1)表达的影响及在动脉粥样硬化发生中的意义。方法 :用细胞培养、免疫荧光定量检测技术观察不同分组条件下白细胞介素- 1α、氧化低密度脂蛋白、高密度脂蛋白对血管内皮细胞VCAM - 1表达的影响 ,并进行比较分析。结果 :IL - 1α组VCAM - 1表达显著高于对照组 ,HDL组VCAM - 1表达显著低于单核细胞粘附状态下对照组 ,而oxLDL组VCAM - 1表达与对照组无显著性差异。单纯培养对照组VCAM - 1表达显著低于单核细胞粘附状态下对照组。结论 :IL - 1、单核细胞粘附可显著增强内皮细胞VCAM - 1表达 ,HDL可显著抑制单核细胞粘附状态下VCAM - 1表达 ,oxLDL对VCAM -
Objective To observe the effects of interleukin-1 (IL-1α), Oxidized low density Lipoprotein (oxLDL), High density Lipoprotein (HDL)and monocyte adhesion on vascular cell adhesive molecule-1 (VCAM-1) expression of endothelial cell. Methods Human umbilical vein endothelial cells (HUVEC) were isolated by pancreatin digestion method and co-cultured with U937 monocyte and IL-1α, oxLDL or HDL respectively. The mean fluorescent intensity of immuno-stained VCAM-1 was determined by ACAS 570. Results (1) IL-1α increased the VCAM-1 expression of HUVEC. (2) oxLDL had no significant effect on the VCAM-1 expression. (3) HDL showed significant inhibition of the VCAM-1 expression induced by IL-1α. (4) Monocyte adhesion enhanced the VCAM-1 expression. Conclusion IL-1α greatly enhances the VCAM-1 expression either in simple culture with HUVEC or in co-culture with HUVEC+U937; HDL may block the VCAM-1 expression induced by IL-1α in monocyte adhesion
