高血压家系AGT基因与细胞因子关系的研究

摘要 目的 研究原发性高血压家系血管紧张素原 (AGT)基因多态性及与细胞因子 (IL 1、IL 6、TNF)的关系。方法 对高血压组 40例和家系对照组 38例用PCR RFLP方法分析血AGT基因型 ,用ELISA方法测定血IL 1、IL 6和TNF浓度。结果 发现AGT2 35TT型在高血压组占 70 % ,在家系对照组占 42 %。血IL 1和TNF浓度在高血压组比家系对照组显著升高 ,在AGT基因2 3 5TT型中比2 35TM型中显著升高。结论 AGT基因M2 35T变异是高血压发病的重要危险因素之一 ,细胞因子IL 1和TNF与高血压发病和AGT基因2 35TT型相关 。 Objective To study the association beween M 235 T allele polymorhpism of angiotensinogen(AGT) gene and cytokine(IL 1,IL 6,TNF) in essential hypertension probands.Methods In 40 hypertensives and 38 proband normotensive controls,polymerase chain reaction combined with restrict enzyme digestion was used to detect the target gene variation,and double antibody sadwich ELISA was used to detect the serum interlukin(IL 1,IL 6),TNF.Results AGT gene T 235 allele frequency(70%) in hypertensives was significantly higher than 42% in proband controls,seum IL 1 and TNF in hypertensives was significantly higher than in controls.At same,IL 1 and TNF in AGT gene 235 TT genotype was significantly higher than in M 235 T genotype.Conclusion AGT gene M 235 T variants may be an importent risk factor for hypertension.IL 1 and TNF may be associated with AGT gene 235 TT genotype and hypertension development.It may play effect in AGT gene variants of hypertensives.
