妊娠妇女血中雌酮和硫酸雌酮的测定

Measurement of Estrone and Its Sulfate During Pregnancy

本文测定了30例妊娠妇女周围静脉血中的雌酮简称E_1(estrone)和硫酸雌酮简称E_1-S(estrone sulfate),采用乙烯醋酸抽提,LH-20 Sephadex柱分离E_1,高效液相色谱仪分离E_1-S。分离E_1-S所用的流动相为50%CH_3CN,50% 0.02M phosphate和2.5g CIMABr。收集的E_1-S片段液经酶解,然后同E_1片段分别用放射免疫法测定。结果是妊娠妇女血中E_1和E_1-S随妊娠周数的增加而增加,其变化关系可分别用以下回归曲线方程描述。妊娠早、中、晚各期的平均值与正常月经周期妇女比较有显著和非常显著差别(P<0.05,P<0.001和P<0.001),反映了妊娠期随着胎儿-胎盘功能的逐步建立,甾体激素除游离型的含量渐增加外,结合型的含量也逐渐增加。本文结果又表明妊娠期随妊娠周数的增加,E_1-S和E_1含量的增加呈非常显著相关性,r=0.95(r_(0.001)=0.46)提示妊娠期随着妊娠周数的增加,游离型和结合型激素的内在调节。为有利于提高胎儿产前监护的正确性,可研究各种游离和结合型的甾体激素。

Peripheral venous blood samples were collected in 30 cases of normal pregnancy. From the samples, estrone and its sulfate were extracted by ethyl acetate; the separation of estrone sulfate was made by HPLC and then solvolized. Estrone and its sulfate fraction were measured by means of PIA. The serum levels of estrone and its sulfate were increased as gestation progressed. The 2 regression formulae were found to be Y_(E_1)=60/1+200.12e^(-0.2006X) Y_(E_1S)=200/1+98.08e^(-0.1867X) The levels of estrone and its sulfate were both significantly than those of normal nonpregnant women (P<0.01) except for those in early pregnancy (P>0.05). The results showed that the free and conjugated steroid were increased during pregnancy. The relation between estrone and its sulfate was very significant (r=095). It was suggested that an inter-control may esist between free and conjugated steriod as gestation progressed.
