摘要
高粱自花受粉后2~6h, 通过花粉管通道将含抗虫基因的重组质粒PAPID-CpTI导入到合子内.所得702粒种子经1000ppm的卡钠霉素初选后,PCR、Southern检测分别得到18株、5株阳性个体,阳性率分别为3.1%、0.86%.结果表明得到了转基因高粱,证明了花粉管通道法可以将抗虫基因导入到高粱体内,获得抗虫的转基因高粱.
The recombinanl plasmids PAP1D-CpTI containing insect-resistant gene are introduced into the zygolyce through the pollen tube pathwa 2-6 hours after the sorghum (sorghum bicolor) are autophilons 702 seeds we get are selected with karnamicin whose concentration is 1000 ppm and the positive rate is 3.10% and 0.86% seperately using PCR detection and southern blotting. The result shows that the transgenic sorghum are produced. It also proves that the transgenic insect-resistant sorghum can be produced by introducing insect-resistant gene into sorghum through the pollen tube pathwa.
