摘要
Id蛋白是碱性螺旋 -环 -螺旋 (bHLH)转录因子之抑制因子。Id - 1蛋白是细胞分化的负性调节因子所以有促进肿瘤发生之功能。本文概述近几年我们研究室在探索Id - 1基因及其功能方面的工作。从动物前列腺癌模型中首次发现Id - 1蛋白之过度表达到人前列腺癌标本中得到验证 ,以及最后在体外培养的前列腺癌细胞株对Id - 1基因之功能研究。实验表明Id - 1是通过抑制P16 RB抑癌基因以及激活有丝分裂原活化蛋白激酶(MAPK)的途径而引起前列腺癌细胞增殖。结果并提示Id -
The protein Id-1 has been suggested to play a positive role in cell proliferation and tumorigenesis of many types of human cancers.we found that the level of Id-1 expression was correlated to Gleason grade of prostate cancer;i.e. low gleason grade,relatively los Id-1 overexpression;high Gleason grade,high Id-1 overexpression.We have further explored under in vitro condition the function of Id-1 gene in prostate cancer cell line,LNCaP,by transfecting a full length human Id-1 gene into LNCaP cells and examined its effects on cell proliferation and cell signaling pathways.Our results showed that Id-1 may stimulate cell proliferation by inactivation the function of p 16 INK4a /pRb and activation of MAPK signaling pathways.Our results suggest for the first time that the function of Id-1 is associated with p16/Rb tumor suppressor gene inactivation and MAPK signaling pathway activation and indicate a possible mechanism in which Id-1 regulates prostate cancer cell growth and tumorigenesis.(Supported by RGC grants to YCW [HKU7186/99M and HKU7314/01M] and Area of Excellence Scheme,UGC of HKSAR,China [Project No.AoE/P-10/01])
