氟化物对大鼠颅骨成骨细胞的细胞周期和细胞凋亡的影响

Effects of fluoride on cell cycle and apoptosis in cultured osteoblasts of rats

目的 :研究氟化物对大鼠颅骨成骨细胞生长、细胞周期和细胞凋亡的影响。方法 :应用酶消化法分离培养大鼠颅骨的成骨细胞 ;AlamarBlue摄入法检测细胞活性 ;流式细胞术检测细胞周期和细胞凋亡。结果 :氟化钠 (NaF)浓度为 0～ 2mmol L、2 4h对细胞活性无影响 ;浓度为 2mmol L时 ,S期细胞数增多 ,G0 G1 期和G2 M期细胞无明显改变。NaF浓度为 4mmol L时 ,细胞活性受抑制 ,S期细胞数明显增多 ,G2 M期细胞明显减少。NaF浓度为 2mmol L和 4mmol L时 ,凋亡百分率明显增加。结论 :过量氟化物可影响大鼠颅骨成骨细胞活性及细胞周期的分布 ,使细胞停滞于S期 。

To study the effects of fluoride on cell growth, cell cycle and apoptosis in cultured osteoblasts of rats. The enzymes digesting method was used to isolate the osteoblasts of rats. The activity of the cells was determined by the percents of reduced AlamarBlue. FCM was used to analyze cell cycle and apoptosis. The results showed that the activity of rat osteoblast was not influenced by NaF at 0 to 2mmol/L concentration after 24 hours incubation. At the concentration of 2mmol/L, the number of cells at S phase was increased. At the concentration of 4mmol/L, NaF increased the number of cells at S phase and at the same time, decreased the number of cells at G 2/M phase. but the number of the cells at G 0/G 1 phase kept unchanged. The percent of apoptosis was increased at the concentration of 2mmol/L and 4mmol/L. Excessive fluoride could affect the cell activity, retarded cell cycle at S phase and induced apoptosis.
