补肾药对复合性免疫损伤大鼠肝纤维化转化生长因子β_1及其受体的影响

The effect of invigorating kidney mixture on transforming growth factor β_1 and its receptor in rats of immunologic injury

目的 研究补肾药对复合性免疫损伤大鼠肝纤维化转化生长因子 β1 (TGFβ1 )及其受体的影响 ,探讨“肝肾同源”理论 ,为慢性肝炎的治疗提供有效的治疗方法和理论依据。方法 3 0只Wistar大鼠随机分为对照组、模型组、补肾组各 1 0只。模型组和补肾组大鼠均腹腔注射 40 %四氯化碳 (CCl4 )花生油溶液和皮下注射 0 .8%小牛血清白蛋白 (BSA)免疫佐剂 5周 ,在损伤第 2日 ,补肾组灌胃给药补肾合剂悬液 ,对照组、模型组给等量水 ,3组均进食普通饲料和水。各组大鼠在 3 6日后分别麻醉状态下称重 ,取肝、肾 ,观察其形态及病理学改变 ;并用ABC法检测肝组织TGFβ1 、TGFβ1 RⅠ、TGFβ1 RⅡ、TGFβ1 RⅢ表达量 ;用原位杂交法检测TGFβ1 mRNA、TGFβ1 RⅠmRNA、TGFβ1 RⅡmRNA水平。结果 ①模型组大鼠生长缓慢 ,肝、肾脏均明显呈缩小 ,与对照组、补肾组比较存在显著性差异 (P <0 .0 5 ) ;补肾组与对照组比较其双肾重存在显著性差异 (P <0 .0 5 )。②大鼠肝组织TGFβ1 及其受体RⅠ、RⅡ、RⅢ表达量和TGFβ1 及其受体RⅠ、RⅡmRNA水平补肾组、模型组与对照组比较均存在显著性差异 (P <0 .0 5 ) ,补肾组与模型组比较TGFβ1 RⅠmRNA水平具有显著性差异(P <0 .0 5 )。③补肾组大鼠病理细胞学改变有明显改善。

Objective To investigate the effect of invigorating kidney mixture on transforming growth factor β 1 (TGF β 1) and its receptor in rats of immunologic injury. Methods 30 Wistar rats were randomly divided into three groups: control group, model group and invigorating kidney group. The immunologic injury model of rats in model group and invigorating kidney group was made by injecting 40% CCl4 peanut oil into abdominal cavity and injecting subcutaneously 0.8% BSA for 5 weeks. The morphological and pathological changes of liver and kidney in three groups were observed, and the TGFβ 1 and its receptors were detected. Results ① The weights of liver and kidney in model group were obviously decreased, as compared with those of control group and invigorating kidney group (P<0.05). ② There were significant differences in TGF β 1 and its receptors of liver tissue between invigorating kidney group and control group and between model group and control group (P<0.05). ③ The pathological changes of tissues in invigorating kidney group were significantly improved. Conclusion The invigorating kidney therapy can protect liver, its mechanism may be related to the inhibition of expression of TGFβ 1 and its receptors, which may relieve liver fibrosis and recover liver function.
