In this report we employed L 929 cell lytic assay for tumor necrosis factor (TNF) to observe the effects of three trace elements(Zinc, Copper and Selenium) on TNF production in normal human peripheral blood mononuclear cells (PBMC). The results showed that zinc and copper were able to augment TNF production in PBMO in vitro. The most optimal concentration of zinc and copper was 0.25 namol/L. They were both able to maintain TNF secretion for as long as 96 hours. The effect of copper was stronger than that of zinc. The results also showed that selenium alone had no effect on TNF production. However, when lipopolysacharide (LPS) was added, they were able to enhance TNF secretion. The present study strongly suggested that zinc, copper and selenium might play an important role in regulating TNF production and their immune regulation function may be mediated by TNF releases.
