中国人的HLA纯合细胞的DR_5分子双相凝胶电泳的多态性分析

摘要 本文应用O'Farrell建立并改进的NEPHGE和SDS-PAGE双相凝胶电泳方法分析了4株中国人HLA纯合细胞的DR_5抗原的多态性。用Iodogen法标记抗原,然后用单抗进行免疫沉淀,得到DR_5抗原后进行双相凝胶电泳,同时还将一株国外参考细胞HVB的DR_5抗原进行电泳并比较其格局。结果表明SMY-129A,SMY-129B属于DR_(w11)而SMY-43A及SMY-124D可能与DR_(w12)有关。 In this experiment we analyzed the DR5 molecular polymorphism of 4 Chinese consanguineous homozygous cell lines(SMY-43A,SMY-124D, SMY-129A, SMY-129B) using the method of 2-dimensional gel electrophoresis developed by O'Farrell. By labelling 125I to the surface antigen of the cell with lodogen, we were able to seperate the DR5 antigen molecules by immunoprecipitation with McAb and to analyze the antigens with 2-dimensional gel electrophorsis. A reference DR5 homozygous cell lines (HVB) was used as control. As a result,we found that the 2D-gel patterns of SMY-129A,SMY-129B were same as or similar to that of HVB, which was identified to be DRwll. The SMY-43A and SMY-124D, however, showed different electrophoresis patterns for DR β chain, a result corresponding to that obtained by polymorphic analysis with POR amplified segments of DR5 genes, in which 43A and 124D were identified to be related to the DRwl2 family.
