乌司他丁对鼠缺血性脑水肿的防治作用 被引量:7

摘要 目的 研究乌司他丁 (Ulinastatin ,UTI)对缺血性脑水肿的防治作用及其机制。方法 应用沙土鼠全脑缺血再灌注模型 ,随机分为 4组 :假手术组 (Ⅰ组 )、缺血组 (Ⅱ组 )、预防组 (Ⅲ组 )和治疗组 (Ⅳ组 ) ,每组 6只。称重法测定脑含水量。测定脑组织一氧化氮 (NO)含量及一氧化氮合酶 (NOS)活性。焦油紫染色光镜观察海马CA1区神经元显微结构。结果 Ⅱ组脑组织含水量明显升高 (P <0 .0 5 ,vsⅠ组 ) ;脑组织NO含量明显上升 ,NOS活性增加 (P <0 .0 1,vsⅠ组 ) ;焦油紫染色光镜显示 :海马CA1区细胞肿胀 ,细胞间隙增大 ,部分细胞核呈不规则固缩、深染。Ⅲ组、Ⅳ组同Ⅱ组相比脑组织含水量明显降低 (P <0 .0 5 ) ,脑组织NO含量减少 ,NOS活性降低 (P <0 .0 1) ;光镜显示 :海马CA1区细胞间隙稍有增大 ,只有个别细胞核固缩 ;Ⅲ组和Ⅳ组无明显差别。结论 乌司他丁具有防治缺血性脑水肿、保护脑细胞的作用 ,作用机制可能与其抑制NOS活性 。 Objective To study the prevention and treating effects of ulinastatin on the experimental ischemic cerebral edema of gerbils and explore the mechanism.Methods The complete cerebral ischemia reperfusion model was used.24 gerbils were randomly assigned to four groups: sham operation group(group Ⅰ),ischemic group(group Ⅱ),prevention group (group Ⅲ),treatment group(group Ⅳ).The content of water within the cerebrum was measured by weighing.The content of nitric oxide(NO) and activity of nitric oxide synthase(NOS) in cerebral tissue were measured.Light microscope was used to study the microstructure of the hippocampal formation.Results Compared with group Ⅰ, the content of water in group Ⅱ elevated significantly in 24 hours (P<0.05); the contents of NO in cerebral tissue elevated significantly,accompanied by increased NOS activity.The following changes in the neurons of CA 1 area of hippocampal formation dyed with cresyl violet may be detected by light microscope: cellular swelling,hyknosis and hyperchromatin.Compared with the group Ⅲ,the content of water in cerebral tissue of group Ⅱ and group Ⅲ decreased significantly (P<0.05),accompanied by decreased contents NO of cerebral tissue as well as decreased NOS activities(P<0.01); Light microscope showed slightly increased intercellular space and a little pyknosis in CA 1 area,with no significant difference between group Ⅱ and group Ⅲ.Conclusion Ulinastatin can prevent, treat cerebral edema and protect the cerebral cells by inhibiting NOS activity and reducing NO.
