摘要
以继代3~4d的长豇豆(Vigna sesquipedalis L.Fruwirth)悬浮细胞系为材料,酶解获得原生质体,在改良的K_8P和MS_1培养基上培养48h后,K_8P培养基中原生质体开始第一次分裂,6~7周形成多细胞团和小愈伤组织。愈伤组织进而在11种不同激素配比和添加不同有机化合物的MS分化培养基中培养,未能获得再生植株。而以MS_1培养基中培养的原生质体不能持续分裂。该结果尚属首次报道。
It is the first report of protoplast calli formation from suspension cells of yardlong cowpea (Vigna sesquipedalis L. Fruwirth). Protoplasts were isolated from suspension cell lines of subculture 3~4d.and cultured in the K_8P and MS_1 media. The protoplasts started to divide after 48 hours in K_8P medium and sustained divisions resulted in mass production of cell colonies and small calli in 6~7 weeks. The calli further grew on the 11 modified MS media with different hormone combinations but no regeneration plants were obtained. The protoplasts did not sustain divisions in MS_1 medium.
