影响大麦花培绿苗和白苗分化的若干因素 被引量:4

FACTORS INFLUENCING THE PRODUCTION OF ALBINO AND GREEN PLANT FROM BARLEY ANTHER CULTURE

摘要 比较了2种低温预处理天数、诱导培养基中添加麦芽糖或蔗糖、添加较高或较低浓度生长素、愈伤组织转移时体积大小、分化培养基中添加不同浓度激素的大麦花培愈伤组织的诱导率、白苗和绿苗分化率。依据试验结果可以认为:25d左右的3~5℃大麦幼穗预处理;诱导培养基中添加3~4mg/L2,4-D、0.5~1.0mg/LNAA、80g/L麦芽糖;分化培养基中添加1.0~2.0mg/L6-BA、0.1~0.5mg/L NAA;愈伤组织体积为2~3mm时转入分化培养基等措施,有利于提高绿苗数量和减少白苗数量。 Two pretreatment-times of low temperature, inducing media with different sugars, concentrations of auxins, regenerating media with different concentrations of hormone and the size of transfered callus were compared for their effects on frequencies of induced callus, regenerated albino or green plant from barley anther culture. According to the comparison, 3 — 5℃ pretreatment of spikes for 25 days, inducing medium with 3~4mg/L 2,4-D, 0. 5~1.0mg/L NAA, 80g/L maltose, regenerating medium with 1. 0~2. 0 mg/L 6-BA, 0.1~0. 5 mg/L NAA and transfer of calli at 2~3mm were suitable for the reduction of albino plant and promotion of green plant.
