NF-кB/Rel蛋白家族是一类参与炎症、免疫、应激、细胞生长及癌症发生等诸多生物学过程中基因调节的重要转录因子。已经发现许多癌症的发生与之相关。本文综述了NF-к B/Rel蛋白在白血病发生及治疗方面的研究进展。NF-кB是正常髓系骨髓细胞转化的调节因子。人类T细胞性白血病病毒Ⅰ型编码的Tax蛋白能通过激活NF-кB而参与人类成人T细胞白血病的发生。一些染色体易位可能影响NF-кB/IкB之间的平衡,从而参与白血病及淋巴瘤的发生。一系列与NF-кB/Rel转录因子相关的白血病化疗药物正在探索之中。
NF-K B and Rel proteins have been found to be important transcription factors which participate in the gene regulation of inflammation, immunity, stress response, cell growth and oncogenesis, NF-K B/Rel family may be involved in the tumorigenesis of many kinds of cancers. The significance of NF-K B/Rel family in the leukemogenesis and the treatment of hematopoietic malignant diseases, mainly leukemia, is reviewed here. NF-K B is a regulator functioning in the transformation of normal myelogenous cells and in regulating the differentiation of myeloid leukemia cells. The Tax protein encoded by HTLV-1 (human T-cell leukemia virus type 1) contributes to human adult T-cell leukemia by activating NF-KlB. Other leukemia virus, e.g. bovine leukemia virus and abelson murine leukemia virus may also induce neoplastic transformation of hematopoietic cells. Some chromosomal translocation may influence the balance of NF-K B/I K B family and play roles in leukemia and lymphoma. New therapeutic strategies targeting NF-K B/Rel are now being developed.
