摘要
研究及应用细胞凋亡相关基因参与肿瘤治疗是基因治疗的一个重要领域。细胞凋亡相关基因中研究得最深入的是p53基因。已经有报道转移野生型p53基因抑制肿瘤细胞的体外实验,或转p53基因抑制动物体内成瘤性的实验。为进一步接近人体内基因治疗,我们采用逆转录病毒载体将p53基因转导至CML细胞,初步的实验结果提示,转导的p53基因能引起白血病细胞生长抑制和凋亡。
The research and application of genes associated with apoptosis are an important field in tumor therapy. To approach human gene therapy in vivo., the wild p53 gene was transduced to CML cells by using retrovirus, the number of the survival cells and percentage of Sub GO were detected and analysed by flow cytometry. The results indicated that transduction of p53 gene could cause growth retardation and apoptosis in CML cells.
