急性白血病患者骨髓基质造血微环境的某些异常<英文>

Some Abnormalities of Stromal Hematopoietic Microenvironment in Bone Marrow of Patients with Acute Leukemia

摘要 为了探索人白血病骨髓造血微环境的异常所在,本实验采用胶体金标记及银增染的免疫电镜方法,检测了急性白血病患者及正常人骨髓长期培养的基质细胞某些生长因子、粘附分子及细胞外基质的表达情况。结果表明,急性白血病患者长期培养的基质细胞表达的GM-CSF(粒-巨噬系集落刺激因子)较对照组高,淋巴细胞归巢相关粘附分子(HCAM),血管内皮细胞粘附分子-1(VCAM-1)和细胞间粘附分子-1(ICAM-1)较对照组低,纤连蛋白(Fn)和Ⅳ型胶原(Col Ⅳ)与对照组无差别。结论提示急性白血病患者骨髓长期培养的基质细胞某些生长因子和粘附分子表达异常。 In order to explore the abnormality of hematopoietic microenvironment in leukemia patients, immunoelec-tron microscopic localization of some growth factors, adhesion molecules and extracellular matrix in microenvironment of leukemia patients and normal controls has been performed by immunocolloidal gold labeling technique. The results show that the expression of GM-CSF by stromal cells from AML patients is higher and HCAM, VCAM-1 and ICAM-1 are lower than that in the controls, and the expression of Fn and Col Ⅳ is as same as controls in HLTBMC. These results suggest that the expression of growth factors and adhesion molecules on stromal cells of leukemia patients is abnormal.
