地氟醚、七氟醚和异氟醚预处理对缺氧/复氧心肌细胞损害的保护作用 被引量:14

Protective effects of desflurane,sevoflurane or isoflurane preconditionins on cultured cardiomyocyte injury induced by anoxia-reoxygenation

摘要 目的 研究地氟醚、七氟醚和异氟醚预处理对心肌细胞缺氧/复氧损害的保护作用。方法 原代培养乳鼠心肌细胞,随机分为对照、单纯缺氧/复氧及 1.5MAC地氟醚、七氟醚和异氟醚预处理5组。实验结束测定乳酸脱氢酶(LDH)和肌酸激酶(CK)活性、细胞存活和凋亡率。结果 与对照组比,单纯缺氧/复氧使LDH、CK和细胞凋亡率升高及细胞存活率显著下降(P<0.01);1.5MAC地氟醚、七氟醚和异氟醚预处理显著减轻LDH、CK和细胞凋亡率升高及细胞存活率下降,其中七氟醚减轻作用最强。结论 地氟醚、七氟醚和异氟醚预处理对心肌细胞缺氧/复氧损害有一定的保护作用,七氟醚的保护作用可能更强。 Objective To study the precoditioning effects of desflurane, sevoflurane and isoflurane on primary cultured myocardial cell injury induced by anoxia-reoxygenation. Methods Ventricular myocytes,cultured in 1640 culture mediums for 4-5 days,were randomly divided into 5 groups: control group receiving no treatment, anoxia-reoxygenation group receiving 2 hours anoxia and 1 hour reoxygenation and 3 preconditioning groups receiving 20 min of preconditioning with 1.5MAC desflurane,sevoflurane or isoflurane before anoxia-reoxygenation respectively. The activities of lactic dehydrogenase (LDH) and creatine kinase (CK),cell viability and apoptosis were measured at the end of the experiment. Results Anoxia-reoxygenation caused dramatic increases of LDH,CK activities and cardiomyocyte apoptosis, and decrease of cell viability compared with control group (358. 17U/L vs 31. 38U/L, 98.83U/L vs 1. 83U/L, 10.19% vs 2.53% and 57.55% vs 90. 77%,respectively). Preconditioning with 1.5MAC desfllurane,sevoflurane or isoflurane significantly attenuated the inoreses of LDH, CK activities and cardiomyocyte apoptosis, and decrease of cell viability caused by anoxia-reoxygenation i. e. 80.83U/L, 34. 83U/L, 88.00U/L; 20.50U/L, 5.33U/L, 15.33U/L ; 6.10 %, 4.77 %, 6. 90%, and 71.34 %, 76.90%, 71.96 % respectively. Conclusion Preconditioning with desflurane,sevoflurane or isoflurane produces a certain protective effects on anoxiareoxygenation injury in primary cultured cardiomyocyte.
