硫唑嘌呤对体外培养中大鼠胰岛细胞功能的影响

摘要 硫唑嘌呤(A)与培养7天新生大鼠胰岛细胞单层培养物孵育16小时,萄萄糖(G)浓度16.7mmol/L时,A 0.5mg/L不影响胰岛素含量(IC)及胰岛素释放(IR),1mg/L只降低IC,2mg/L既降低IC也降低IR;G浓度5.6mmol/L时,A2mg/L也不影响IC及IR。A2mg/L (G 16.7mmol/L)降低IC及IR在去除A16小时后能完全恢复。 Effect of azathioprine (Az) on insulin content (IC) and insulin release (IR) in neonatal rat islet cell monolayer cultures was investigated. At 16.7 mmol/L of glucose, IC and IR were not affected after 16-hour incubation in the medium containing 0.5 mg/L of Az. IC was reduced at 1 mg/L of Az, and both IC and IR were reduced at 2 mg/L of Az. However, the inhibitory effect fully recovered after a further 16-hour incubation in the Az-free medium. At 5.6 mmol/L of glucose, Az (2mg/L) did not affect IC and IR.
