帕金森病猴模型的神经生化学研究 被引量:1

摘要 应用HPLC-EC法和SPECT在体显像检测由MPTP制备的双侧和偏侧帕金森病猴模型的神经递质含量和DA受体活性。结果发现:(1)6只双侧和4只偏侧帕金森病猴模型的尾状核、壳核和黑质DA及其代谢产物显著降低,DOPAC/DA和HVA/DA比值明显增高;(2)2只偏侧帕金森病猴模型病变侧的D_2 DA受体活性在症状初期无改变,症状明显时发生超敏。研究表明,帕金森病猴模型的神经递质含量变化和DA受体活性改变符合帕金森病患者的神经生化学改变。 The contents of neurotransmitters and the activity of dopamine rceptor in parkinsonian monkeys rendered by 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) were determined by high performance liquid chromatography (HPLC) and single photon emission computed tomography (SPECT) imaging, respectively. Marked depletion of DA, DOPAC and HVA and significant elevation of DOPAC/DA and HVA/DA ratios were found in the caudate nucleus, putamen and substantia nigra of the monkeys with generalized parkinsonism and hemiparkinsonism (lesioned side). SPECT scans in hemiparkinsonism monkeys showed no change of D_2 dopamine receptor activity in the lesioned striatum at the emergence of parkinsoism symptoms, and showed denervation supersensitivity when the symptoms became evident. It is concluded that these neurobiochemical changes occurred in parkinsonian monkeys rendered by MPTM are almost similar to those in human Parkinson's disease.
