硫唑嘌呤对大鼠葡萄糖代谢和β细胞功能的影响

摘要 本研究观察了硫唑嘌呤对大鼠葡萄糖代谢和β细胞功能的影响。给Sprague-Dawley大鼠腹腔注射硫唑嘌呤5或15mg·kg^(-1)·d^(-1)8天,与对照组比较,结果不影响动物体重、血浆葡萄糖、胰岛素及果糖胺水平(P>0.05);但明显降低胰腺胰岛素含量和/或胰岛素释放(P<0.01)和<0.05)。 The effect of azathioprine on glucose metabolism and β-cell function in rats was observed. Azathioprine 5 or 15 mg·kg_(-l)·d_(-1) was given intraperitoneally to Sprague-Dawley rats once daily for 8 days. It was found that animal weight, fasting plasma glucose, insulin and fructosamine were not effected in comparison with those of controls (P<0.05). On the 9th day, the pancreas of animals was removed, minced and incubated in a medium` of glucose 16.7mmol/L for 1 hour or homogenized and extracted in acid ethanol for 24 hours at 4℃. Azathioprine given at a dose 5mg.kg_(-1)·d_(-1), the insulin content and insulin release decreased to 77.13% and to 79.57% of controls respectively (P<0.05 ond P>0.05). When azathioprine was given at a dose of 15 mg·kg_(-1)·d_(-1), the insulin content and insulin release decreased to 54.60% and to 73.75% of controls respectively (P<0.01 and P<0.05). These findings confirmed that azathioprine had adverse effect on pancreatic islet β-cell function.
