摘要
在建立Bcl-2高表达细胞及其对照细胞株基础上,用无细胞体系研究了细胞质在顺铂诱导HL-60细胞凋亡中的调控作用。实验结果显示,凋亡细胞胞质提取物可引起提取的正常细胞核的固缩及染色质边缘优,类似于完整的细胞凋亡的改变。Bcl-2高表达细胞的胞质提取物具有抗凋亡能力。上述结果说明引起凋亡的物质及对抗凋亡的物质在细胞质中都存在,说明细胞质在顺铂诱导的HL-60细胞凋亡中起着较重要的作用。
On the basis of establishment of Bcl-2 overexpression cells and control cells, the role of cytoplasm in apopto-tic HL-60 cells induced by cisplatin was studied in a cell-free system. The results showed that the extracts of cytoplasm of apoptotic cells could induce condensation and margination of chromatin along the nuclear envelope in normal cells, just like what happened in intact cells, while that of Bcl-2 overexpression cells could be more apop-tosis-resistant. The results suggset that the materials inducing and resisting to apoptosis are existed in cytoplasm, and the cytoplasm plays an important role in apoptosis induced by cisplatin in HL-60 cells.
