摘要
[目的 ]探讨体外细胞培养制备微核模型的环磷酰胺的适宜剂量 ,为以后的抗突变物质的功能评价提供依据 .[方法 ]进行体外淋巴细胞培养的微核实验 .[结果 ]不同浓度 (2 5,5 0 ,10 0 ,2 0 0mg/L)的环磷酰胺对淋巴细胞导致的微核率和微核分布有不同的影响 ,较低浓度时具有致微核率升高的性质 ,较高浓度的环磷酰胺除具有致突变作用外 ,还能显著地抑制细胞的分裂和生长 .[结论 ] 5
OBJECTIVE?To study the proper concentration of cyclophosphamide (CP)-induced micronuclei model in order to support the lat er research on anti-mutation.?METHODS?The micronuclei experiment was carried out by lymphocytes cell culture in vitro .?RESULTS?Different concentrations (2 5,5 0,10 0,20 0? mg/L) of CP could result in different rate and distribute of CP-induced micronucle i, and at lower concentration, the CP could cause increase of micronuclei, while at higher concentration, it could cause both mutation and prohibition of ce ll growth.?CONCLUSION?5 0?mg/L of the CP is the proper conce ntration for inducing mutation by cell culture.
