摘要 [目的 ]为棘阿米巴感染的病原诊断、形态学观察及进一步的研究建立棘阿米巴的实验室培养方法 .[方法 ]将含有阿米巴的 1g土壤置于敷有大肠杆菌的 15mg/L琼脂培养基上 ( 2 5℃ )培养 1周 ,取单个棘阿米巴包囊纯培养 ,以 0 1mol/L盐酸进行杀菌处理后 ,用PYG培养液进行无菌培养 .[结果 ]从第 3日起可在培养基上观察到滋养体 ,第 5日开始部分形成包囊 ,棘阿米巴滋养体形态呈不规则 ,具有特征性棘状伪足 ,颗粒状胞质内有较多空泡 .包囊具有双层囊壁 ,外囊皱缩 ,内囊呈四边形或波浪状 .[结论 OBJECTIVE?To establish the methods of isola tion and culture of the Acanthamoeba sp.? METHODS ?One g ram of soil was incubated on an 15?mg/L agar plate which had been covered with E.coli and incubated for 1 week. The cysts of a clone was treated 0 1?mol /L HCl and incubated in PYG.?RESULTS?The trophozoites was dete cted from third day, and the cysts was formed particularly in fifth day. The mor phology of trophozoites was irregular with special acanthopodia with a slower tr anslation. The cyst wall is two layer with wave like or quadrangular endocyst and moderately undulated exocyst.?CONCLUSION?The expe riment methods of isolation and culture of the Acanthamoeba have been established.
