摘要 用DNS法测木聚糖酶活力 ,分析测定条件对测定结果的影响。结果表明 ,在不同的条件下测定酶活力会得到不同的木聚糖酶活力测定值。其中 ,酶液用量、酶解反应的时间对测定结果的影响较大 ,酶液稀释度、DNS显色时间、DNS的用量 ,对测定结果也有一定的影响。测定木聚糖酶活力的适宜条件为 :酶液量 1%木聚糖液量 1 9(V V) ;酶解反应时间 10min ;DNS用量 2 - 2 .5ml;DNS显色时间 2 - 5min。 To analyse the effection of different conditions of measurement on the measuring results,when using DNS method measure the activity of xylanase.The results show that the activities of enyzyme will appear to be different under different conditions,Among of these conditions,the quantity of enzyme and the time of the reaction have more effect on the results. While the dilute rate of xylanase,the time DNS takes to appear its colour,the quantity of DNS also have something to do with the results.The optimum condition of determination xylanase activity was studied.The proportion of xylanase and 1% xylan is 1:9(V/V);the reaction time is 10 minute; the dose of DNS is 2-2.5 ml,and the time DNS taking to appear its colour is 2-5 minutes.
