摘要
目的 :建立芒果叶中芒果苷的含量测定方法 ;测定不同产地和月份芒果叶中芒果苷的含量。方法 :采用RP HPLC法 ,内标法定量 ,ShimpackCLC ODS柱分离 ,流动相 :甲醇 0 .0 5mol·L- 1H3PO4(三乙胺调pH为 3 .5)65∶13 4 ,检测波长 2 58nm ,柱温为室温 ,流速 1mL·min- 1。从广西南宁市、钦州市和田阳县分别采集芒果叶供试品 ;测定南宁市 1~ 12月份同一株芒果树的芒果叶中芒果苷的含量。结果 :所建立的RP HPLC含测方法平均回收率 99.2 % ,RSD =1.0 5% (n =5)。不同产地和月份的芒果叶中芒果苷的含量存在差异。结论 :所建立的RP HPLC含测方法灵敏度高 ,准确 ,快速 ,专属性强 ,重现性好。南宁市和田阳县芒果叶中芒果苷的含量相差不大 ,均明显比钦州的高。 7~ 10月份的芒果苷含量较高 ,最高的为 9月 ,最低的为
Objective: To develop a new method for det er mination of Mangiferin in the leafs of Folium mangiferae. By this new method , Mangiferin in F. mangiferae sampled in different months and in different r egions was determinated. Method: A RP-HPLC method was set up, using Shim pack CLC-ODS column, methanol-0.05 mol·L -1 H 3PO 4 (65 ∶134, pH 3.5) as mobile phase, with 258 nm of detection wave, at room tem perature, 1 mL·min -1 . F. mangiferae sampled in Nanning, Qinzhou an d Tianyang, Guangxi province and sampled respectively in January to December we re determinated. Result: The average recovery of the RP-H PLC was 99.2%, RSD=1.05%(n=5). The content of Mangiferin in F. mangifera e was statistically different due to different sample-regions or sample-time . Conclusion: This RP-HPLC method is simple, specific and exact. The contents of Mangiferin in the leafs of F. mangiferae sample in Nanning and Tianyang were statistically similar, but higher than that in Qinzho u. The contents of Mangiferin in the leafs of F. mangiferae sampled in July to October were higher than that in the other months. The content in September was the highest, the content in February was the lowest.
