摘要
目的 探讨低分子肝素对大鼠肾缺血—再灌注损伤的防治作用。方法 建立肾缺血—再灌注损伤大鼠模型 ,实验动物随机分为正常对照组和低分子肝素治疗组及非治疗组。观察大鼠治疗前后肾组织中P 选择素表达、组织病理学与肾功能的改变 ,以及凝血指标和凝白酶原时间 (PT)、白陶土部分凝血活酶时间 (KPTT)的变化。结果 缺血—再灌注后大鼠肾脏P 选择素表达增高 ,出现明显的组织病理学改变及肾功能损害。低分子肝素治疗组肾组织中P 选择素表达受抑 ,病理学改变与肾功能损害减轻 ,且对PT和KPTT无明显影响。结论 低分子肝素对肾缺血—再灌注损伤具有防治作用 ,且对凝血指标PT和KPTT无明显影响。
Objective To investigate the effect of low molecular weight heparins(LMWH) on renal ischemia reperfusion injury in rats. Methods Using a rat model of 60 minutes renal ischemia followed by 1,3,6 and 24 hours reperfusion,animals were randomly divided into following groups:the sham operated controls,ischemic group receiving only normal saline,and treatment group receiving LMWH at a dose of 1 5 mg/kg at 5 minutes before reperfusion.P selectin expression was observed in renal tissues with immunohistochemisal method. Results P selectin expression, blood urea nitrogen (BUN) and creatinine (Cr) levels were significantly increased after 60 minutes ischemia and 24 hours reperfusion,showing a notable histopathological change and impairment of renal function,but P selectin expression of LMWH group was inhibited,showing a less pathological change and a lighter renal impairment,and not affecting the prothrombin time (PT) and kaolin partial thromboplastin time (KPTT).Conclusion The results suggest that LMWH have preventive and curative effect on renal ischemia reperfusion injury,without affecting the clotting indices PT and KPTT.
