TRAP-SYBR-Green染色法检测大鼠睾丸组织内端粒酶活性的实验研究

摘要 目的 研究不同年龄SD大鼠睾丸组织内端粒酶活性表达及其意义。方法 应用TRAP SYBR Green染色法 ,对各年龄组健康雄性SD大鼠睾丸组织中端粒酶活性进行了检测 ,并结合光镜、电镜观察对端粒酶活性在不同年龄SD大鼠睾丸组织内的表达状况进行了研究。结果 从出生后第 1d直到第 2 7个月 ,各年龄组雄性SD大鼠的睾丸组织中均可检测到端粒酶活性的表达 (阳性率 10 0 % )。出生后第 9~ 39dSD大鼠睾丸中可检测到高水平的端粒酶活性。A型精原细胞表达高水平的端粒酶活性。结论 SD大鼠睾丸组织中终生表达端粒酶活性 ,A型精原细胞作为雄性生殖系统的干细胞 。 Objective To study the telomerase activity in the testis of SD rats and its significance.Methods Telomerase activity in the testis of 102 different age male SD rats was detected by TRAP SYBR Green staining and a human carcinoma cell line (HL 60) as a standard. More study has been done with the aid of microscopy and electron microscopy. Results The positive percentage of telomerase activity in the testis was 100 % from 1 day old to 27 month old SD rats. There was a high level of telomerase activity in type A spermatogonia. Conclusion The telomerase activity in the testis of SD rats is all their life. High level of telomerase activity may play an important role in the proliferation and differentiation of type A spermatogonia.
