雌、孕激素对卵巢癌细胞株体外生长的影响

摘要 目的 观察 17β 雌二醇 (E2 )、E2 与黄体酮 (P)联合应用对人上皮性卵巢癌细胞株A2 780体外生长的影响。方法 分对照组和实验组 ,用四甲基偶氮唑蓝 (MTT)比色法计算细胞生存率 ,用流式细胞术测定细胞周期。结果 ①浓度为 0 1~ 5 0nmol/L的E2 可抑制卵巢癌细胞株的生长 ;加入浓度为 5 0nmol/L的P后这种抑制作用增强。②浓度为 0 1~ 5 0nmol/L的E2 对卵巢癌细胞株细胞周期有影响 ;G0 /G1期细胞由 5 7 11%增加到 71 99% ,S期细胞由 32 39%减少到 2 0 5 5 % ,加入浓度为 5 0nmol/L的P后 ,细胞周期进一步发生变化 ,G0 /G1期细胞由5 7 11%增加到 77 39% ,S期细胞由 32 39%减少到 15 80 %。结论 雌激素对卵巢癌细胞株A2 780的增殖具有抑制作用 ,并呈现剂量 效应关系。雌。 Objective To study the effect of 17β estradiol and progesterone on the growth of the epithelial ovarian cancer A2780 lines in vitro . Methods Cell proliferative capacity was measured by 3 2,5 dipheny tetrazolium blue colorimetric assay. Cell cycle was detected by flow cytometry. Results Estradiol of 0 1-0 5 nmol/L inhibited the growth of A2780 cell. After addition of progesterone of 50 nmol/L, this inhibitory effect was enhanced. Estradiol of 0 1-0 5 nmol/L changed A2780 cell cycle with the cell rate of stage G 0/G 1 being increased from 57 11 % to 71 99 % and the cell rate of stage S being decreased from 32 39 % to 20 55 %. After progesterone of 50 nmol/L was added, the cell cycle changed further with the cell rate of stage G 0/G 1 being increased from 57 11 % to 77 39 % and the cell rate of stage S being decreased from 32 39 % to 15 8 %.Conclusion Estrogen inhibited A2780 cell growth in a dose dependent manner. Estrogen combined with progesterone can enhance this inhibitory effect.
