摘要
目的 探讨蛛网膜下腔出血 (Subarachnoidhemorrhage,SAH)后 ,脑底动脉神经肽Y(NeuropeptideY ,NPY)能神经纤维变化与脑血管痉挛发生与发展的关系 ,并为深入阐明SAH的发病机制提供形态学资料。方法 Wistar大鼠 42只随机分为 3组 :(1)正常组 6只 ;(2 )假手术组 6只 ;(3)手术组 :30只。按SAH后 12h、1d、2d、3d、7d再分为 5小组 ,每组各 6只。应用免疫组织化学ABC法 ,对正常组、假手术组和手术组大鼠的脑底动脉NPY能神经纤维进行染色并观察。结果 各组大鼠脑底血管均可见棕褐色的细线、弯曲状的免疫反应阳性纤维 ,手术组 (SAH后 12h、1d、2d)与正常组相比较 ,纤维密度明显的减少 (P <0 .0 5 ) ,表明手术组与正常组脑底动脉的NPY能神经纤维密度有较显著的差异。结论 NPY能神经在SAH鼠脑血管痉挛的发生和发展中起重要的作用。
Objective To explore the relationship between changes of neuropeptide Y nerves in cerebral artery and cerebral vasospasm after the subarachnoid hemorrhage(SAH)in the rats,and to offer new clinic morphologic data.Methods Forty-two Wistar rats were randomly divided into three groups.(1)normal group(n=6),(2)sham-operted group(n=6),(3)experimental groups(n=30).Again,the experimental groups were divided into five groups based on time points:at 12 hours、1、2、3、7 days after subarachnoid hemorrhage,the number of animal in each group was 6.The density of neuropeptide Y innervation in the cerebral arteries of the normal group,sham-operted group,and the experimental groups was studied with immunoperoxidase ABC technique.Results The immunopositive fibers with the brown linear structures were observed in the cerebral arteries in the rats of normal group,sham-operted group.Neuropeptide Y nerve fibers were less dense in cerebral arteries of SAH than those of normal rats(P<0.05).Conclusion It suggested that the neurooeotide Y nerves mihgt play an important role in the maintenance and the development of cerebral vasospasm after SAH.
