p16和p53抑癌基因在肾母细胞瘤中的表达及与预后关系

The Relationship of Expression of Gene p16 and p53 with Biological Behavior and Prognosis of Wilms Tumor

目的 探讨p16、p5 3抑癌基因在肾母细胞瘤中的表达及与临床预后的关系。 方法 应用免疫组织化学方法S -P和ABC法检测p16和 p5 3基因的表达。 结果 2 0例肾母细胞瘤标本p16蛋白染色阳性 7例 ,阳性率为35 %;其中上皮型 10例 ,阳性 4例 (占 40 %) ;间叶型 5例 ,阳性 1例 (占 2 0 %) ;胚胎型 3例 ,阳性 1例 (占 33%) ;混合型 2例 ,阳性 1例 (占 5 0 %)。p5 3蛋白阳性染色 5例 ,阳性率为 2 5 %。其中上皮型及混合型 12例中 ,p5 3阳性 2例 (占 16 .7%) ;而间叶型及胚胎型 8例中 ,阳性 3例 (占 37.5 %)。结论 对肾母细胞瘤患者同时进行p16和 p5 3检测更能准确地判断其病理性质和预后。

Objective To study the tumor supressor genes p16 and p53 which express in Wilms' tumor and to explore the relation between the expression of the two genes and the pathological grade,histological type and the prognosis.Methods Twenty species from patients with nephroblastoma were analyzed by the immunohistochemical techniques of S-P and ABC.Results p16 protein were positive in 7 cases(35%),while p53 protein were positive in 5 cases(25%).The positive rate showed a significant relation with the pathological type and prognosis.Furthemore,p16 was more expressed in epithelial type and mixed type of Wilms tumor,which indicated the prognosis was better,whereas p53 protein was found more in blastemal and stromal predominant tumors with a poor prognosis.Conclusion Combination detection of p16 and p53 is helpful to investigate the biological behavior of nephroblastoma and predict the prognosis.
