丹参对自发性高血压大鼠心肌细胞凋亡及基因蛋白表达的影响 被引量:9

The effect of Salvia Miltiorrhiza on Cardiomyocyte Apoptosis and Gene Protein Expression in Spontaneously Hypertensive Rats

摘要 目的探讨丹参对自发性高血压大鼠(SHR)心肌细胞凋亡及基因蛋白表达的影响。方法18只引种繁殖的雄性SHR随机分为3组,每组6只:⑴基础组:育至第8周处死;⑵丹参组:从第8周给药,丹参粉针剂1g/kg.d,腹腔注射,第18周处死;⑶对照组:用相同容量蒸馏水腹腔注射,余同丹参组。心肌细胞凋亡的检测采用TdT介导的原位末端缺口标记法(TUNEL),Bcl-2基因的蛋白表达检测采用S-P免疫组化法。比较3组间心肌细胞凋亡指数、Bcl-2蛋白阳性表达指数的差异,分析两指标间的相关性。结果与基础组比较,对照组心肌凋亡细胞明显增多(P<0.05),而Bcl-2蛋白阳性表达指数显著降低(P<0.05);丹参组较对照组心肌凋亡细胞明显减少(P<0.05),Bcl-2蛋白阳性表达指数显著升高(P<0.05),与基础组无差异。心肌细胞凋亡指数与Bcl-2蛋白阳性表达指数呈明显的负相关性(P<0.01)。结论SHR心肌细胞凋亡具有周龄的依赖性,中药丹参能显著减少SHR心肌细胞凋亡,具有明显上调Bcl-2基因蛋白表达的作用。 Objective To observe the effect of Salivia Miltiorrhiza on cardiomyocyte apoptosis and gene protein expression in spontaneously hyperten-sive rats(SHR).Methods18male SHR were di-vided in3group at random(n=6):⑴the base group,which were killed in the8th week;⑵the Salvia Miltiorrhiza group,which Salvia Miltiorrhiza(1g /kg.d)were injected to intraperitoneally from the8th week to the18th week;⑶the control group,which the same volume of vehicle were injected in the same manner.The TUNEL method was employed to eval-uate cardiomyocyte apoptosis,and the immunohis to chemistry method was employed to detect the pro-tein expression of Bcl-2.Results Compared with the base group,cardiomyocyte apoptosis increased markedly in control group (P<0.05),the positive ex-pression index of Bcl-2protein decreased signifi-cantly (P<0.05),while the cardiomyocyte apoptosis decreased and the positive expression index of Bcl-2protein increased in the Salvia Miltiorrhiza group compared with the control group(P<0.05).The cardiomyocyte apoptosis index showed negative cor-relationship with Bcl-2protein expression.Con-clusions The study show an age -dependent in-crease in apoptosis in the cardiac tissues of SHR.The chinese medicinal herb Salvia Miltiorrhiza can decrease the cardiomyocyte apoptosis and upregulat-ed the protein expression Bcl-2gene in SHR.
