摘要
本文利用KSOM、TCM-199+20%人血清和KSOM+20%人血清培养绵羊体外受精胚胎,以研究KSOM作为绵羊胚胎体外培养无血清培养液的可能性。实验结果说明,KSOM支持绵羊胚胎早期发育至囊胚期,而在KSOM中添加血清后,胚胎可发育至孵化囊胚,而且孵化囊胚率高于常用的TCM-199+血清(24%vs 7%)(p<0.05)。说明KSOM可以用于绵羊胚胎早期发育的无血清培养液。
KSOM, TCM-199+20% human serum and KSOM+20% human serum was used as serum free medium in ovine embryo development. The results indicated that ovine embryos could develop to blastocyst in KSOM, and could develop to hatched blastocyst in TCM-199+20% human serum and KSOM+20% human serum. The rate of hatched blastocyst in KSOM+20% human serum was higher than that in TCM-199+20% human serum (24% vs 7% ) (P<0.05). This paper suggested that KSOM can be used as serum free medium for ovine embryo development in vitro.
