免疫磁珠细胞术分选脐血CD_(34)^+细胞方法的改良

摘要 本研究采用免疫磁珠细胞术分选脐血CD+34 细胞 ,并对其方法进行改良。按CD34+细胞MACSReagents按试剂盒的操作方法 (旧方法 )分选 9例脐血标本 ,用改良后的方法 (新方法 )分选 1 0例脐血标本 ,每例标本均进行了两次分选。结果 ,旧方法于第一、二次分选CD34+细胞的分离率分别为 4 0 0± 5 0 %和 86 3± 5 2 % ,新方法于第一、二次份选中的分离率分别为 88 4± 1 5 9%和 92 4± 6 5%。旧方法与新方法在第一次分选CD34+细胞的分离率比较有显著性差异 (P <0 0 1 ) ,在第二次分选的比较无差异 (P >0 0 5) ,新方法第一、二次分选结果的比较无差异 (P >0 0 5)。结果表明 :采用新方法在第一次分选CD34+细胞的分离率就已达到旧方法第二次分选的效果 。 To study the method of enrichment of cod blood CD 34 + cells by immunoaffinity selection. Mehtods: 9 cord blood units were sorted by the method of CD 34 + cells MACS Reagents Kit(old method), 10 units were sorted by our improved method (new method). All units were sorted twice. Results: The purity of CD 34 + cells sorted one time was 40.0±5.0%, purity of those sorted second time was (86.3±5.2%), purity of CD 34 + cells sorted one time and second time was (88.4±15.9%) and (92.4±6.5%), respectively. There was significant variance in the purity of CD 34 + cells sorted one time between by old method and by new method (P<0.001). There was no significant variance in the purity of CD 34 + cells sorted second time between two methods (P>0.05), that was also true in the purity of CD 34 + cells sorted by new method between one time and second time (P>0.05). Conclusions: The purity of CD 34 + cells sorted one time by new method equals to that sorted second time by old method. It means simple operation and money saving.
