氨溴索注射液对茶碱在家兔体内药动学的影响

Effect of ambroxol injection on the pharmacokinetics of theophylline in rabbits

目的 :研究氨溴索注射液对茶碱在家兔体内药动学的影响。方法 :采用自身对照法 ,用高效液相色谱法 (HPLC)测定合用氨溴索后 ,茶碱不同时间间隔血药浓度 ,采用 3P97程序进行模型判别及参数计算。结果 :合用氨溴索注射液后茶碱的t1/ 2 β由 (3.2± 0 .8)延长至 (4.0± 1.1)h(P <0 .0 5 ) ;AUC由 (75 .0± 2 2 .4 )增至 (98.2± 2 7.9)mg·h·L-1(P <0 .0 1) ;CL由 (0 .2 0± 0 .0 8)下降至 (0 .15± 0 .0 4 )L·h-1(P <0 .0 5 )。结论 :氨溴索注射液与茶碱在家兔体内合用可影响茶碱的t1/ 2 β、AUC、CL。

OBJECTIVE To study the effect of ambroxol injection on the pharmacokinetics of theophylline in rabbits.METHODS Healthy rabbits were given theophylline with either water or ambroxol. Blood samples were collected at different time points. Plasma was assayed for thephylline by HPLC and various pharmacokinetic parameters were analyzed by 3P97 software.RESULTS As compared to controls,the t _ 1/2β ,AUC and CL of theophylline were significantly changed in the rabbits with ambroxol injection coadministration. The t _ 1/2β were increased to ( 4.0 ± 1.1 )h from ( 3.2 ± 0.8 )h( P < 0.05 ),the AUC were increased to ( 98.2 ± 27.9 )mg·h·L -1 from ( 75.0 ± 22.4 )mg·h·L -1 ( P < 0.01 ),otherwise the CL were decreased to ( 0.15 ± 0.04 )L·h -1 from ( 0.20 ± 0.08 )L·h -1 ( P < 0.05 ). CONCLUSIONS The results indicate that ambroxol injection may affect the t _ 1/2β , AUC and CL of theophylline.
