针刺手厥阴心包经穴对心肌缺血再灌注损伤凋亡调控基因表达的影响

Effect of Acupuncture the Points of Hand-Jueyin on the Genes Expression of Apoptosis in Myocardial Ischemia and Reperfusion Injury

目的 :观察针刺手厥阴经穴对心肌缺血再灌注损伤过程中细胞凋亡调控基因bcl 2、bax的影响。方法 :电针大鼠心包经穴位 2 0min后 ,结扎左冠状动脉前降支 4 0min ,心电图 (ECG)监测 ,再电针穴位 2 0min ,松扎 ,恢复灌流 6 0min ,摘取心脏 ,免疫组化方法分析凋亡细胞基因的表达。结果 :模型组bcl 2蛋白表达的阳性率较低 ,bax蛋白表达的阳性率明显增高 ,针刺心包经穴组与模型组比较均有非常显著意义 (P <0 0 1)。结论 :针刺手厥阴经穴对细胞凋亡的不同调控基因有促进或抑制的作用 ,由此可以减轻由于凋亡细胞所导致的心肌损伤。

Purpose:To explore the effect of acupuncture the points of Hand Jueyin on the genes expression of bcl 2 and bax during the process of ischemia and reperfusion injury.Methods:Acupuncture are given at the points of Hand Jueyin for 20min,followed by ligation of anterior descending branch of the left coronary artery and 40min later,acupuncture the points for 20min again,reperfusion for 60min,with ECG monitoring.Then,cardiac tissue was taken and the genes expression of bcl 2 and bax was observed by immunohistochemical method.Results:In the model group,The gene expression of bcl 2 is few,otherwise the gene expression of bax is more.Compared with acupuncture groups,the changes are significantly( P <0\^01).Conclusion:Acupuncture the points of Hand Jueyin can obviously influence the genes expression of bcl 2 and bax,reduce the injury degree during the process of ischemia and reperfusion injury.
