摘要
对棉花染色体三种制片方法进行了比较研究,结果表明:涂片法方便快速,一张载玻片有较多可观察的细胞,但容易发生染色体丢失,因此不适合染色体计数和组型分析。滴片法制备的染色体标本,染色体的清晰度和分散度都较理想,但是如果酶解法,成本较高,如果用盐酸水解,技术难度比压片法更大。压片法制备的染色体标本,完全符合计数和组型分析的要求,用盐酸水解的难度要求相对较小,因此是棉花染体制备最为实用的方法。本文同时对每一种制片方法关键技术进行了探讨。
The articale studies on three methods' comparasion of getting cotton's chromosome. They indicate: 1. smearing - method is convenient, but cotton s chromosomes are frequently missed. 2. dropping - method has the best experiment result, but it is expensive or needs hard skills. 3. crushing - method is the best practical method of getting cotton' s chromosome in the whole.
