摘要 目的 :探讨并建立金属 β 内酰胺酶IMP基因的PCR方法。 方法 :临床分离的病原菌用全自动微生物分析系统鉴定 ,纸片扩散法进行抗生素敏感试验 ,采用 2 巯基丙酸络合金属离子的纸片扩散法来检测金属酶表型 ,PCR法检测金属酶IMP基因。结果 :2 7株耐亚胺培南细菌金属酶表型共检出产金属酶菌株 8株 ,其中铜绿假单胞菌 7株、嗜麦芽窄食单胞菌 1株 ;7株表型产金属酶铜绿假单胞菌IMP基因扩增阳性 ,其他扩增均阴性。结论 :该方法是一种灵敏度高。 Objective: To explore and establish PCR method for detection of IMP g ene of metallo-β-lactamases. Methods: Pathogenic bacteria were identified by VIT EK-AMS. Antimicrobial susceptibility tests were performed by K-B method and me tal lo-β-lactamases produced by bacteria was detected by using 2-mercaptopropion ic acid in the M-H agar, and IMP gene of metallo-β-lactamases was detected by P CR method.Results: eight strains of metallo-β-lactamases- producing bacteria, which included 7 strains of Pseudomonas aeruginosa and 1 strain of Stenotrophomonas maltophili a,were detected in 27 strains of Imipenem-resistant bacteria.Amplification of I M P gene was positive in 7 strains of metallo-β-lactamases-producing Pseudomo nas aeruginosa,and negtive in other strains.Conclusion: PCR was a highly se nsitive and specified method for detection of IMP gene of metallo-β-lactamase s.
