摘要
将pSC101上的par区域克隆到载体pUC9上,得重组质粒pEC302。将pUC9和pEC302分别转入E.coli HB 101,在无机培养基M_9中试验两者的稳定性,结果发现E.coli HB101(pUCg)传代培养40代后质粒几乎全部丢失,而E.coli HB101(pEC302)的质粒全部存在。并且发现质粒的稳定性与宿主菌的recA基因有关。
The par region of pSC101 was cloned into pUC9. Then pEC302 was obtained, The stability of pEC302 and pUC9 in E.coli HB101 was tested on M_9 medium to determine the effect of par region on the plasmid stability. After forty generations, pUC9 was almost lost, while pEC302 was 100% maintained in host cells.
