摘要
着床前的兔胚泡液经ConA-Sepharose亲和柱层析和Sephadex G-100凝胶过滤柱层析,得到大分子量(ConA GP 1)和小分子量(ConA GP 2)糖蛋白两个组份。作者进一步观察了这两组份对孕兔子宫内膜蛋白质和前列腺素F 2α(PGF 2 α)合成与分泌的影响。以~3H-Leu、~3H-GlcNH 2,~3H-Gal和~3H-Fuc分别作为蛋白质、糖蛋白上糖链合成的前体,用RIA法测定PGF 2α浓度。结果发现胚泡液ConA GP两组份均选择性地调节于宫内膜蛋白质的合成与分泌和糖蛋白上糖链的装配。ConA GP1增加子宫内膜~3H-GlcNH 2标记糖蛋白的合成,减少它的分泌,高浓度时抑制。~3H-Leu标记蛋白质的合成。ConA GP 2除具有与ConA GP 1一致的对~3H-GlcNH 2标记糖蛋白的作用外,还增加子宫内膜~3H-Gal标记糖蛋白的合成,减少它的分泌。高浓度的ConA GP 2抑制子宫内膜。~3H-Leu标记蛋白质的合成与分泌。两种组份对~3H-Fuc标记糖蛋白的影响不显著。结果还表明,两种组份均显著减少子宫内膜PGF 2α的释放。显然,着床前兔胚泡分泌糖蛋白参与了孕兔子宫内膜蛋白质、糖蛋白中糖链和PGF 2α的代谢调节。
Glycoprotein of rabbit blastocyst fluid was isolated by ConA-Sepharose affinity chromatography and Sephadex G-100 gel filtration. Two fractions, ConAGP 1 (large MW) and ConAGP 2 (small MW) were collected. The effects of ConAGF 1 and ConAGP 2 on synthesis and secretion of protein, assembly of glycoprotein and PGF_(2α) secret icn were examined. ~3H-Leu, ~3H-GlcNH_2, ~3H-Gal and ~3H-Fuc were used as precursors of protein and glycolinkage synthesis. PGF_(2α) concentration was determine by RIA. The results showed that ConAGP 1 increased ~3H-GlcNH_2 labelled glycoprotein synthesis, decreased its secretion, and also inhibited ~3H-leu labelled protein synthesis of endometrium. ConAGP 2 had the same effect on ~3H-GlcNH_2 labelled glycoprotein as ConAGP 1. Moreover, it increased ~3H-Gal labelled glycoprotein synthesis and decreased its secretion and ~3H-Leu labelled protein synthesis and secretion. Neither ConAGP 1 nor ConAGP 2 had any effect on ~3H-Fuc labelled glycoprotein. The result also indicated that ConAGP 1 and ConAGP 2 inhibited PGF_(2α) secretion of endometrium. It demonstrated that glycoprotein of rabbit blastocyst was implicated in the regulation of protein, glycoprotein and PGF_(2α) metabolism of endometrium.
