摘要
目的 探讨Flt3L对恒河猴外周血树突状细胞前体 (pDC1和pDC2 )的免疫扩增作用。方法 健康猴艾滋病毒SIV阴性的恒河猴每日给予rhFlt3L 10 0 μg/kg体重皮下注射 ,于第 11天在氯胺硐全麻下采集静脉血 ,用Ficoll Hypaque梯度离心法提取外周血单核细胞 ,利用与恒河猴有交叉反应的人单克隆抗体以及三色流式细胞仪分离 pDC1和 pDC2 ,并与未经Flt3L作用的 pDC1和pDC2 在数量和细胞表现型上进行对比。结果 经Flt3L扩增的恒河猴外周血 pDC1和 pDC2 数量分别增加 7倍和 4倍 ,且细胞表现型和形态不发生改变。结论 Flt3L对恒河猴外周血pDC1和pDC2 具有免疫扩增作用。
Objective To study the immune amplification of peripheral blood pDC 1 and pDC 2 in rhesus monkey by Flt3L.Methods rhFlt3L (100 μg/kg every day) were subcutaneously injected into healthy,SIV-negative rhesus monkeys (M.mulatta).After 11 days,all animals were anesthetized with Ketamine and peripheral blood samples were collected.Ficoll-Hypaque density method was used to separate the mononuclear cells of peripheral blood.pDC 1 and pDC 2 were isolated by rare-event cytometric flow analysis using human monoclonal antibodies cross reactive with rhesus monkey and compared with those not treated with rhFlt3L in quantity and phenotypes.Results Populations of pDC 1 and pDC 2 were increased 7 fold and 4 fold respectively by Flt3L administration with the stabilization in quantity and phenotypes.Conclusion Flt3L increased pDC 1 and pDC 2 of rhesus monkey peripheral blood.
