摘要
建立测定人血浆中塞曲司特的高效液相色谱法。采用 C1 8柱 ,流动相为甲醇 - 0 .0 2 m ol/ L KH2 PO4 溶液 (75∶2 5 ) ,流速 1.2 m l/ min,柱温 37°C,检测波长 2 6 8nm。内标为辛伐他汀。线性范围 0 .0 5~ 10μg/ m l(r=0 .9998) ,用乙酸乙酯 -环己烷提取 ,提取回收率 97.9%~ 10 0 .7%。
A HPLC method for determination of seratrodast in human plasma was established. Simvastatin was used as the internal standard and ethyl acetate-cyclohexane was used as extractive solution. A C_ 18 column was used with the mobile phase of methanol-0.02 mol/L KH_2PO_4 solution (75∶25), at detection wavelength of 268 nm. The flow rate was 1.2 ml/min at column temperature 37 °C . The calibration curve was linear over concentration range of 0.05~10 μg/ml( r=0.9998 ). The extraction recovery was 97.9%~100.7%
