摘要
目的 :探讨复制 SD大鼠抗 Thy1.1系膜增生性肾炎实验模型的方法 ,观察其临床病理特点和肾小球系膜区细胞增殖核抗原 (PCNA)、凋亡调节基因 Bcl- 2的表达。方法 :用 SD大鼠胸腺细胞免疫新西兰白兔 ,制备兔抗大鼠胸腺细胞免疫血清 (Anti- Thy1.1serum,ATS) ,再将 ATS经尾静脉注射给 SD大鼠 ,诱导其发生系膜增生性肾炎(Ms PGN)。每日检测尿蛋白 ,用 HE和 PASM染色法观察肾小球系膜增生程度 ,用免疫组化 L SAB法检测系膜区 PCNA及 Bcl- 2的表达。结果 :ATS效价可达 1∶ 32 0 0 ;诱导大鼠 Ms PGN的 ATS最适用量为 :0 .0 1m L/ g体重 ;大鼠 Ms PGN的临床特点为大量蛋白尿 ;病理特点为肾小球系膜细胞增生 ;增生期系膜区 PCNA、Bcl- 2的表达增高。结论 :用 ATS可诱导典型的、稳定的大鼠 Ms PGN实验模型 ;免疫反应和细胞凋亡受抑制参与了肾小球系膜增生的发病机制。
Objective:To explore the way to reproduce a rat model with mesangial proliferative glomerulonephritis(MsPGN) induced by anti Thy 1.1,and to investigate its clinicopathologic characteristice and the expression of PCNA and Bcl 2 in the mesangial area.Methods:Anti Thy1.1 serum(ATS) was produced in New Zealand white rabbits by immunization with SD rat thymocytes,and then injected intravenously to induce SD rat model with MsPGN.Proteinuria was measured everyday,the degree of mesangial proliferation was observed by HE,PASM and Masson’s stains,and the expression of PCNA and Bcl 2 in the mesangial area was detected by LSAB method.Results:The titre of ATS could amount to 1∶3 200,and the optimal dosage to induce rat model was 0.01 mL/g·wt.The rat model was characterized by heavy proteinuria,mesangial proliferation,and enhanced expression of PCNA and Bcl 2 in the mesangial area.Conclusion:A stable and typical rat model with MsPGN can be successfully induced by ATS.The immune response and inhibition of apoptosis of mesangial cells may play an important role in the mesangial proliferation of the rat model with MsPGN.
