摘要
目的 :观察抗冻剂渗透压对深低温冻存角膜内皮细胞活性的影响。方法 :牛角膜随机分两组 ,分别使用二甲基亚砜 (DMSO) +2 0 %人血清白蛋白 (HSA)即对照组及二甲基亚砜 (DMSO) +CPTES即实验组溶液配制的抗冻剂采用二步平衡法冻存 ,观察两种方法对复温后角膜内皮细胞密度、存活率、细胞均匀性及六角形细胞比例等的影响。结果 :使用 CPTES溶液配制抗冻剂冻存的细胞密度及存活率与对照组比较差异均无显著性 ,但细胞均匀性与六角形细胞比例均较对照组优。结论 :CPTES溶液配制抗冻剂有助于改善冻存的细胞活性。
Objective:To investigate effects of osmotic stress of cryoprotectants on viability of cryopreservative corneal endothelial cells.Methods:New born bovine corneas were divided into two groups randomly,and then cryopreserved with the simplified “two step” method.Human serum albumin(HSA) and corneal potassium TES(CPTES) were chosen as cryoprotectant vehicles in group Ⅰ and group Ⅱ,respectively.The indexes,including cellular density,survival rate,polymegathism and pleomorphism,were compared respectively after thawing between two groups.Results:There were no significant differences on cellular density and survival rate between two groups,while group Ⅱ was superior to group Ⅰ on both polymegathism and pleomorphism.Conclusion:CPTES solution,an important ingredient of cryoprotectant,is helpful in improving the viability of cryopreservative corneal endothelial cells.
