中药对老龄大鼠脑缺血再灌注单胺神经递质的影响 被引量:3

Effect of TCM Recipes on Monoamine Nervous Transmitter in the Aged Rats with Brain Ischemia Reperfusion

摘要 从多巴胺 (DA)和去甲肾上腺素 (NE)及肾上腺素 (E)的变化方面研究青年大鼠和老龄大鼠脑缺血再灌注损伤及研究益元活血丹和大黄及血栓心脉宁对老龄大鼠脑缺血再灌注损伤防护作用的机制 ,为防治脑梗塞提供依据。青年 (5月龄 )大鼠分为模型组和正常对照组 ,老龄大鼠 (2 0月龄以上 )分为模型组和对照组、尼莫地平组、益元活血丹组、大黄组、血栓心脉宁组 ,观察各组大鼠全脑缺血 30 min再灌注 6 0 min后脑组织病理和 NE、DA、E含量的变化。结果 ,青年和老龄模型组大鼠脑组织均出现病理改变 ,交感—肾上腺系统兴奋增强 ,老龄模型组大鼠较青年模型组大鼠严重。用药各组老龄大鼠脑组织损伤和交感—肾上腺系统兴奋减低 ,大黄组和益元活血丹组的交感—肾上腺系统兴奋减低明显。交感—肾上腺系统兴奋增强与脑缺血再灌注损伤有关 ,老龄大鼠较青年大鼠严重 ;各组用药对老龄大鼠脑缺血再灌注脑组织损伤的保护作用可能与其抑制交感—肾上腺系统兴奋增强有关 ,其中以大黄和益元活血丹的作用较为全面而显著。 Objective:To study the brain ischemia reperfusion injury in the young and the aged rats and the protective effect of Yiyuanhuoxuedan(YHD),rhubarb and Xueshuanxinmaining (XXN) on brain ischemia reperfusion injury in the aged rats from the changes of Dopamine(DA),Noradrenalin(NE) and Epinephrine(E) in order to provide theoretical basic for prevention and cure of cerebral infarction.Method:Young rats(5 months)were divided into model group and normal control group,aged rats(more than 20 months)were divided into model group,control group,nimodipine group,YHD group,rhubarb group,XXN group.The pathological change in brain tissue and the NE,DA and E content were measured in the rats with 60 minute reperfusion after 30 minute brain ischemia.Results:The pathological change was found in the young model's and the aged model's,the excitability of sympathetic-adrenal system was improved,this change in the aged model's was serious compared with the young model's.The brain injury and excitability of sympathetic-adrenal system were alleviated in the aged rats in every taking medicine group,and the decrease of the excitability of sympathetic-adrenal system was evident in the rhubarb's and the YHD's.Conclusion:The enhance of the excitability of sympathetic-adrenal system was correlated with brain ischemia reperfusion injury in the aged rats.This injury in the aged rats was more serious than that in the young rats.The TCM drugs of every group in this trial could alleviate the injury of brain in the aged rats via restraining the excitability of sympathetic-adrenal system,YHD and rhubarb's effect was remarkable and comprehensive among them.
