摘要
建立了高效液相色谱法测定人血清中氟西汀的分析方法。用岛津LC-6A型高效液相色谱仪,色谱柱为ZorbaxODS(150 mm×4.6 mm,5μm);流动相为:甲醇-水-1 mol/L醋酸铵-5 mol/L氨水(850:150:10:3);检测波长为265 nm;流量为0.81 ml/min;柱温40℃;灵敏度0.005AUFS;纸速2 mm/min。线性范围为100~2500 ng/ml(r=0.9984),检测限为20 ng/ml。平均回收率为(96.21±2.93)%。日内平均相对偏差为3.1%(n=15),日间平均相对偏差为4.5%(n=5)。
A method for the determination of fluoxetine level in serum with HPLC was established. The conditions were Zorbax ODS (150 mm×4. 6 mm, 5 μm) column; 0. 8 ml/min flow solution, methanol-water-1 mol/L ammonium acetate-5 mol/L ammoia (850 : 150 : 10 : 3); UV-265 nm detector. The linear range was 100 ng/ml to 2500 rig/ml (r = 0. 9984) and the detection limit was 20 ng/ ml. The average recovery was (96. 21 ± 2. 93)% and the coefficients of variation were 3. 1%(n=15) and 4. 5% (n=15) for within-day and day-to-day tests respectively.
