摘要 目的 :研究大环内酯类药物对铜绿假单胞菌生物膜的影响。方法 :用高分子膜片制备铜绿假单胞菌生物膜 ,计数滤膜菌落与扫描电镜观察 ,观察大环内酯类药物对铜绿假单胞菌生物膜形成前后的影响及对已形成生物膜的清除作用。结果 :2mg/L阿奇霉素、2mg/L红霉素可抑制生物膜的形成 ,生物膜菌落计数由对照 1 0 .0× 1 0 9cfu/cm2 ,分别降至 1 .9× 1 0 3cfu/cm2 、5 .0× 1 0 3cfu/cm2 。麦迪霉素作用不明显 ;单用阿奇霉素、红霉素及麦迪霉素不能清除已形成的生物膜 ,但阿奇霉素、红霉素与环丙沙星协同作用 ,可使已形成生物膜中细菌下降至 0 .9%~ 2 .2 %。电镜观察与上述结果一致。结论 :红霉素、阿奇霉素能抑制铜绿假单胞菌生物膜的形成 ,增强环丙沙星对生物膜的渗透性 ,增强其杀灭生物膜内细菌的作用 ,清除生物膜 ; Purpose:To investigate the effects of macrolides on Pseudomonasmol aeruginosa biofilms. Methods: Pseudomonas aeruginosa biofilm was prepared using high malecular membrane, the number of colonies in biofilm were counted, then observed by scanning electron microscopy. Results: The number of colonies in biofilms were reduced from 10×10 9 cfu/cm 2 in controlled biofilmm to 1.9×10 3 cfu/cm 2 treated with 2?mg/L azithromycin, and to 5.0×10 3 cfu/cm 2 treated with 2?mg/L erythromycin. The number of colonies in biofilms treated with azithromycin, erythromycin or medemycin alone had no significant effect on biofilm, however when treated with azithromycin or erythromycin in combination with ciprofloxacin, the number of colonies decreased to 0.9%-2.2% of the original. No significant change was observed when medemycin combined with ciprofloxacin. Similar results were obtained when observed with scanning electron microcopy conclusions. Conclusions:These results indicate that azithromycin and erythromycin can inhibit the formation of biofilm by P. aeruginosa ; enhance the bactericidal action of ciprofloxacin on bacteria within the biofilm. Midecamycin has no such effect.
