大鼠大脑中动脉局灶性缺血再灌注单胺类递质的变化 被引量:2

Variation of monoamine neurotransmitters in the focal cerebral ischemia-reperfusion injury for rats

摘要 目的 :观察大鼠大脑中动脉 (MCA)局灶性缺血再灌注期间脑内单胺类递质的变化。方法 :Wistar大鼠 6 0只 ,随机分为 :①正常组 ,②假手术组 ,③缺血 1h组 ,④缺血 2h组 ,⑤缺血 2h再灌 1h组 ,⑥缺血 2h再灌 2h组。采用线栓法制作大鼠MCA局灶性缺血再灌注模型 ,用荧光分光光度计测定大脑皮层和纹状体多巴胺 (DA)、去甲肾上腺素 (NE)、5 -羟色胺 (5 -HT)含量。结果 :与②组比较 ,③组大脑皮层和纹状体DA含量明显降低 (P <0 .0 5 ) ;④、⑤组大脑皮层和纹状体DA、NE、5 -HT含量均显著降低 (P <0 .0 1 ) ;⑥组DA、NE、5 -HT含量与②组相比差异无统计学意义 (P >0 .0 5 )。结论 :MCA缺血再灌注时 ,大鼠大脑皮层和纹状体DA、NE、5 -HT含量显著降低 ,降低程度与缺血时间长短有关 ,其中DA降低较明显。提示DA、NE、5 Aim:To observe the variation of monoamine neurotransmitters in the middle cerebral artery occlusion (MCAO) rats.Methods:Sixty Wistar rats were randomly allocated into 6 groups, that was normal group, sham operation group,1 h ischemia group,2 h ischemia group,2 h ischemia and 1 h reperfusion group,and 2 h ischemia and 2 h reperfusion group.The models of MCAO were made by occluding the MCA with thread.NE,DA,and 5-HT were measured by fluorospectrophotometry.Results:Compared with sham operation group,cortex and striatum DA contents of 1 h ischemia group decreased significantly ( P <0.05); NE,DA,and 5-HT contents of 2 h ischemia group and 2 h ischemia and 1 h reperfusion group decreased significantly ( P <0.01);but difference in NE,DA and 5-HT contents between 2 h ischemia and 2 h reperfusion group and sham operation group were not significant ( P >0.05).Conlusion:NE,DA, and 5-HT contents in cortex and striatum decreased significantly after reperfusion, and the degree of reduction is related to the time of ischemia-reperfusion.DA decreased more significantly.NE,DA, and 5-HT involve in the pathophysiologic process of focal cerebral ischemia-reperfusion injury.
