应用SOS显色试验研究消毒剂的遗传毒性 被引量:1

摘要 在遗传毒性的检测中,细菌测试系统已为世界许多国家采用,因为它具有快速、简便等应用上的优点,并可提供遗传毒性的基本机制及其后果。细菌是含DNA的最简单的细胞,它拥有对DNA损伤剂作出反应的机制。在E.Coli中,已知有一套SOS反应功能(17个基因构成的SOS反应网络)在DNA损伤剂处理后作出表达。因此,可通过监测细菌细胞中SOS反应的方法以检测受试物的遗传毒性。近年来,已为多数学者应用于检测食品、有害气体、金属及其无机化合物、烷化剂和抗肿瘤药物的遗传毒性。本文即应用此法检测了七类14种消毒剂的遗传毒性。 The genotoxicity of 14 disinfectants, including formaldehyde, glutaraldehyde, ethylene oxide, sodium dichloroisocyanurate, peracetic acid, hydrogen peroxide, ethanol, isopropyl alcohol, carbolic acid, lysol, bromogeraminum, sodium dichloroisocyanurate mixture, fungicide and Qianglisu, was studied using the SOS chromotest that is a rapid System for detecting genotoxic agents by monitoring SOS responses. In this system, the SOS function induced by genotoxicants is detected by a colorimetric measurement of β-galactosidase in E. coli PQ 37&PQ35 carrying a sfiA::lac Z fused gene. Results from study show that most of disinfectants tested in this experiment have no higher β-galactosidase activity. Among these 14 disinfectants, only ethylene oxide and Qianglisu gave positive results. Comparison of the results from SOS chromotest and Ames test revealed a strinking concordance.Several practical advantages of SOS chromotest were discussed.
